A physical framework for the peach genome

We are constructing a physical framework for peach, which is a model fruit tree for the Rosaceae species. Currently the physical framework is composed of 1,128 contigs containing 8,508 BAC clones. Conservative estimates indicated these contigs cover at least 53% of the peach genome. Based on core genetic marker hybridizations, 168 contigs were anchored onto an integrated physical/genetic map. The 47 positioned contigs were localized on linkage group 1, which represents the most marker-saturated chromosome of peach. To verify the physical framework, we used 673 ESTs of the peach unigene set (PP_LE) and 64 cDNA (LF) clones, which were incorporated into the framework by random fingerprinting. At this juncture, more than 12% of BAC clones in the peach FPC database have hybridization hits (i.e. core genetic markers, ESTs, cDNAs).
