Aptamer-based nanofluidics for the molecular detection in ultra-small volume

Abstract

Molecular detection in samples with sub-picoliter (picoliter, pL =10−12 L) volumes is a challenge impeding the development of several fields such as single cell studies at the single molecule level. As a proof-of-concept study, here we proposed the aptamer-based nanofluidic device for the detection in samples with femtoliter (fL, 10−15 L) volumes. Platelet-derived growth factor BB (PDGF-BB) and its aptamers were used. The aptamers were modified onto the gold nanopattern surfaces inside nanochannels, and could recognize targets with high selectivity. This study proved that the aptamer-based nanofluidics is promising in bioanalysis area especially in the single-cell omics studies.
