General serum and muscle proteins were starned wrth 0.7% Buffalo Black NBR In 7% acetrc acid for 3-4 h. Destarnrng was accomplished usrng successive changes of 7% acetrc acid lsozyme starnrng technrques are lrsted In Table 1 Both parental species, the hybnd and an equal mixture of the parental species were used during each electrophoretrc run Saturated wrcks were used for applicatton of samples onto the rsoelectnc focusing gels For gradrent gels, 4 ~1o f serum and 20 ~1o f muscle homogenate were Inserted Into sample wells Hrstochemrcal stainrng procedures followed those described rn the lrterature 129-331 ( Table 1) Acknowledgements - Thus research was supported by the Unrted States Forest Servrce through grant 53.56Al-O-00372 Freld assrstance from the following rndivrduals IS gratefully acknowledged’ t. N2 - Campostoma anomalum, Rhinichthys cataractae and their F1 hybrids were examined electrophoretically for 44 enzymatic loci, general muscle, and serum proteins. Of the 44 loci scored, acid phosphatase (ACP-B), alkaline phosphatase (AKP-A), esterase (EST-B), α-glycerophosphate dehydrogenase (GPD-A), malate dehydrogenase (MDH-A) and phosphoglucomutase (PGM-A) showed hybrid inheritance patterns. Serum proteins also demonstrated additive inheritance patterns, comprising 15 serum proteins in the parents and 18 in the F1 hybrid. Banding patterns for all mixtures of parental species were identical to those observed in the hybrid.
