N2 - Pyrpose. The effects of PEDF on rat retinal cells were studied, Methods. Primary cultures of lewborn albino rat retinas were treated with PEDF (500 ng/ml) at 2 DIV. At 9 DIV, cultures were studied by phase contrast nicroscopy, SEM, TEM and immune-fluorescence techniques. Results. PEDF-treated cultures showed more numerous and larger pigment epithelial cells, with several cell-cell contacts, and a higher amount of more mature pigment granules than in controls. Conclusions. PEDF promotes survival and differentiation of pigment epithelial cells. These findings show that PEDF may be one of the factors of the autocrine-paracrine system whch operates in the pigment epithelium-photoreceptor complex.
