Abstract
(Cell 186, 786–802.e1–e15; February 16, 2023) In our original published article, we performed western blots and imaging of induced motor neurons (iMNs) to elucidate the mechanism through which PIKFYVE inhibition causes secretion and clearance of aggregation-prone proteins such as phosphorylated TDP-43 (pTDP-43). Since publication, we have become aware of 3 errors in figure legends or images that we are now correcting.(1)In Figures 3F and S3F, we intentionally showed the 55 kDa pellet TUJ1 bands from the same blot in both figures. We also noticed that the full blot for the 55 kDa pellet TUJ1 bands in Figure 3F in Data S1 was flipped 180 degrees about the vertical axis by accident. To correct, we are now indicating that the western blots in Figures 3F and S3F are from the same cell samples and that we chose to only include some of the blots in Figure 3F for clarity and due to space constraints in the main figure. Therefore, we included the 55 kDa pellet TUJ1 bands again in Figure S3F to improve clarity. We have revised the figure legend and figure labeling in Figure S3F to explicitly communicate this. We also corrected this in Data S1.(2)In Figures 3B and 3J, we intentionally showed that the 55 kDa pellet TUJ1 bands as duplicates given the same samples were used for Figures 3B and 3J. For clarity, we split the data into two panels because it flowed better with the pTDP-43 analysis being presented in Figure 3J after showing evidence of exosomal secretion of autophagosomal markers in Figures 3C–3I. In this correction, we are including the 55 kDa pellet TUJ1 bands from Figure 3B again in Figure 3J for clarity, which is scientifically appropriate because this is not a loading control, but rather a control for the relative number of neurons in each sample. In this correction, we also explicitly indicate in Figure 3J, the legend for Figure 3J, and Data S1 that the 55 kDa pellet TUJ1 bands in Figures 3B and 3J were from the same blot.(3)In Figure 3J and the respective legend, we mistakenly indicated that the 46 kDa exosome TSG101 bands were from the same blot as in Figure 3B. To correct this, we have revised the labeling on Figure 3J, the legend for Figure 3J, and Data S1 to indicate that the 46 kDa exosome TSG101 bands were from the same samples as in Figure 3B rather than the same blot.(4)In Figure S4G, we accidentally used the images from columns 2 and 3 (control cells) in columns 6 and 7 (C9-ALS cells). In addition, the labels for columns 2 and 3 were reversed, and so were the labels for columns 6 and 7. We have now corrected these errors. Importantly, the published quantification in Figure S4G is still accurate and all conclusions remain the same for this experiment.
