Tetranucleotide markers based on cosmids from human chromosome 13 library

New markers of the human chromosome 13 were generated using tetranucleotide microsatellites. The cosmids with GATG and GACT motifs isolated earlier from the chromosome 13 library [1] were analyzed with Southern blotting and located subregionally using fluorescence in situ hybridization. We have subcloned 16 cosmids located at the longer arm of the chromosome 13. Tetranucleotide repeats were detected in 14 sequences of microsatellites analyzed with standard and specific primers. Six markers were generated using PCR. The polymorphism of these loci was studied for groups of 30-140 unrelated individuals. Allele and genotype frequencies were analyzed for the three most informative markers (4-6 alleles, heterozigosity 0.64-0.75). The highly informative locus D13S699 appeared useful for practical paternity tests.
